Summary

Migraine is a common neurological disorder causing recurrent headaches. Female migraine patients often report less headache severity and frequency during late pregnancy. However, the reasons behind this phenomenon are not well understood. This study created a chronic migraine rat model for pregnant subjects. Researchers found that oxytocin levels increased in the paraventricular nucleus and nucleus of the solitary tract during late pregnancy. Viral tracing showed that oxytocinergic projections travel from the paraventricular nucleus to the nucleus of the solitary tract. Intranasal oxytocin administration significantly lowered CGRP expression and pro-inflammatory cytokine levels. Additionally, it alleviated hyperalgesic behaviors in these models. The protective effects of oxytocin were linked to downregulating the extracellular matrix-receptor interaction signaling pathway. This research highlights how elevated oxytocin during pregnancy may help mitigate migraine-related neuroinflammation and pain. It opens the door for future oxytocin-focused migraine treatments.
